Neurobasal™-A Medium

Neurobasal™-A Medium is a basal medium formulated to meet the special cell culture requirements of post-natal and adult brain neuronal cells when used with GIBCO™ B-27™ Supplement. Neurobasal™-A Medium can be used to grow neuronal cells from hippocampus, cortex and other regions of the brain. Neurobasal™-A Medium allows for both long and short term maintenance of homogeneous populations of neuronal cells without the need of an astrocyte feeder layer.

Add Your Own Supplements
All Neurobasal™ media contain no L-glutamine, L-glutamic acid, or aspartic acid. Neurobasal™ media must be supplemented with 0.5 mM L-glutamine or GlutaMAX™-I Supplement and either a serum-free supplement (such as B-27™ Supplement or N-2 Supplement), or with serum. For initial plating, you should add 25μM glutamic acid.

Quality Control Testing
All Neurobasal™ media are tested for pH, osmolality, endotoxin, and the absence of bacterial and fungal contamination. The osmolality of Neurobasal™-A was demonstrated to be optimal for postnatal and adult CNS neurons.

Neurobasal™-A Medium contains Phenol Red. If Phenol Red will interfere with your applications, we also provide Neurobasal™-A Medium Minus Phenol Red.

Neurobasal™-A Medium is optimized for post-natal and adult brain neurons. For growing pre-natal and embryonic neurons, we provide Neurobasal™ Medium.

Can Neurobasal Medium alone or Neurobasal Medium supplemented with B-27 supplement be frozen to extend product shelf life?

No, it is not recommended to freeze media due to the potential of precipitates forming upon thaw. Inorganic salts and amino acids in the formulation may come out of solution when exposed to temperature fluctuations. These precipitates will not go back into solution easily once formed.

What supplements should I use with Neurobasal or Neurobasal-A medium?

Neurobasal and Neurobasal-A media (for postnatal and adult neurons) allow for long-term maintenance of neuronal cells without the need for an astrocyte feeder layer. These media should be supplemented with either serum or a serum-free supplement, plus 0.5mM L-glutamine. B-27 supplement is a serum-free supplement that comes as a 50X concentrate in a 10ml volume. This is enough supplement for 500ml of media. Fetal, postnatal, and adult neural cultures can be grown in the appropriate Neurobasal medium supplemented with B-27 supplement .

We also have two other supplements. One is called G-5 and is for growth and expression of glial cells (normal and tumor) of astrocytic phenotype. This comes in a 1ml size, at a 100X concentration. The other supplement is called N-2 and is for growth and expression of post-mitotic neurons and tumor cells of neuronal phenotype. This comes in a 5ml size, at a 100X concentration.

Can you provide references for growing adult neurons?

Instructions for growing adult neurons can be found in the Neurobasal-A Medium product insert. You can also refer to the following publication for growing adult neurons:

Should I use Neurobasal-A Medium with B27 and b-FGF for postnatal neurons?

We do not know at what age postnatal neurons require FGF and a higher osmolality medium, or how this could vary based on the species from which the neural tissue is isolated. Without this knowledge, we generally recommend switching to the Neurobasal-A Medium for isolates from animals above 1 week of age. You will need to determine if the Neurobasal-A Medium is optimal for neurons you have from one day old rats.

Further, the addition of bFGF can increase the viability of neurons of any age. Therefore, caution needs to be made in impure neuronal cell culture as bFGF can increase the proliferation of non-neuronal cells. We recommend a pilot experiment to test the impure culture in the presence and absence of bFGF.

What should I add to Neurobasal-A Medium to grow adult neurons?

We recommend adding B27 and b-FGF to Neurobasal-A Medium to grow adult neurons.

What did you add to Neurobasal-A Medium to increase osmolality?

4 g/L of NaCl has been added to the Neurobasal-A Medium formulation to increase osmolality.

Which media system should I use for primary culture of P0- or P1-derived mixed hippocampal cells?

We recommend using Neurobasal Plus Medium with B-27 Plus Supplement for long-term culture of mixed hippocampal cells. For short-term culture of a pure population of hippocampal neurons, we would recommend using Neurobasal-A Medium with B-27 Supplement.

Which supplement can I add to Neurobasal Medium to culture my neural cells?

As a general guideline, we recommend using Gibco B-27 Plus Supplement to culture neural stem cells, hippocampal, and other CNS neurons.

- For neuroblastomas, or post-mitotic neurons from both PNS and CNS, Gibco N-2 Supplement can be used.
- For primary glial cells (astrocytes) or tumor cell lines of astrocytic phenotype (astrocytes and gliomas), or oligodendrocytes, Gibco G-5 Supplement can be used.

What is the difference between Gibco Neurobasal Medium and Gibco Neurobasal-A Medium?

Neurobasal Medium is optimized for prenatal and fetal neurons. Neurobasal-A Medium is optimized for growing postnatal and adult brain neurons. These two media differ only in osmolality.
