Description
- The ReadyProbes Tissue Autofluorescence Quenching Kit minimizes unwanted autofluorescence in FFPE and frozen tissues due to aldehyde fixation, red blood cells, and structural elements such as collagen and elastin.
- If the target signal is low enough, then this background signal can interfere with the interpretation of immunohistochemistry (IHC), immunocytochemistry (ICC), immunofluorescence(IF), and in situ hybridization (ISH) assays.
- The ReadyProbes Tissue Autofluorescence Quenching Kit minimizes most kinds of autofluorescence, with the exception of autofluorescence from lipofuschin granules.
- The ReadyProbes Tissue Autofluorescence Quenching Kit contains three components that are combined just before use. Combine components A, B, and C in a 1:1:1 ratio.
- Mix components A and B together before adding component C. The resulting solution is then applied to the tissue for 2–5 minutes at room temperature.
- This is done after the blocking, primary antibody, and secondary antibody steps, but before application of any counterstains like nuclear stains.

No. The ReadyProbes Tissue Autofluorescence Quenching Kit minimizes autofluorescence created by aldehyde fixation, red blood cell autofluorescence, and autofluorescence of collagen and elastin. It will not quench natural pigments or other autofluorescent entities.
For the ReadyProbes Tissue Autofluorescence Quenching Kit to work, the components need to be mixed in the recommended order. Component A needs to be mixed with Component B, followed by mixing in Component C.
Once the 3 components are mixed together, we recommend using this mixture within 1 hour. The mixture should not be stored beyond 1 hour.
