Pierce Protease Inhibitor Tablets protect proteins from proteolytic degradation through inhibition of the serine-proteases, cysteine-proteases, aspartic acid-proteases and aminopeptidases that are typically present in cellular lysate samples. The tablets contain AEBSF, aprotinin, bestatin, E-64, leupeptin and pepstatin A and are available in formulations with or without EDTA, a metalloproteinase inhibitor. The EDTA-free formulation has added compatibility with IMAC purification methods and 2D gel electrophoresis. Stable for storage at 4°C for up to 12 months, each large tablet is sufficient for 50mL of solution and each mini tablet is sufficient for 10mL of solution. Both sizes and formulations of tablet are suitable for reagent preparation prior to protein extraction from tissue and cultured cells.

Pierce Protease Inhibitor Tablets (Cat. No. A32963) dissolve quickly to form a clear solution. We recommend dissolving 1 tablet in 50 mL of buffer or lysate. The protease inhibitor formulation is compatible with most detergent-based cell lysis reagents, including Pierce Cell Lysis solutions. Pierce Protease Inhibitor Tablets do not contain phosphatase inhibitors. If necessary, phosphatase inhibitors, such as Pierce Phosphatase Inhibitor Mini Tablets (Cat. No. A32957) can be added along with the protease inhibitor tablets.

Can I use just part of the Pierce Protease Inhibitor XL Capsules, EDTA-Free and save part of the capsule for later use?

We do not recommend “splitting” the capsule into smaller portions. Researchers using less than 500 mL should choose a different format (liquid or tablet) of protease inhibitors.

However, it is important to avoid using EDTA when the downstream application involves metal chelation affinity purification or if the target protein requires metals ions for proper function.
