Thermo Scientific™ Pierce Glycoprotein Isolation Kits use ConA and WGA lectin affinity resins and optimized buffers to efficiently isolate glycoproteins from serum, tissue and cultured cell lysates, thus enabling downstream analysis.
Glycosylation is a post-translational modification that plays an important role in biological functions, including immune regulation, inflammation, cell-to-cell adhesion and cell signaling. Asparagine (N-linked) and serine/threonine residues (O-linked) are glycosylated during passage through the endoplasmic reticulum and golgi apparatus in eukaryotic and prokaryotic (i.e., Archaea and Eubacteria) systems. Glycoconjugates are important for immune regulation, inflammation, cell-to-cell adhesion and contact inhibition, cell signaling, protection against proteolytic degradation, and other biological processes.

The ConA kit (Cat. No. 89804), which has concanavalin A immobilized on the resin, recognizes alpha-linked mannose, and to a lesser extent, terminal glucose residues. The WGA kit (Cat. No. 89805), which has wheat germ agglutinin immobilized on the resin, recognizes N-acetyl glucosamine (GlcNAc) groups and sialic acid.
Enrichment of specific proteins or protein complexes can most easily be accomplished by using target-specific immobilized antibody or PTM specific affinity binding.
