The MagMAX™ Cell-Free DNA Isolation Kit is designed for enrichment of circulating cell-free DNA (cfDNA) and optimized for use with biological samples such as serum and plasma. The kit is based on MagMAX™ magnetic-bead technology, enabling reproducible recovery of high-quality DNA that is suitable for a broad range of applications, including real-time PCR, digital PCR, and next-generation sequencing.

Scalable isolation of circulating nucleic acid from plasma, serum, and urine
cfDNA recovery is challenging due to the limited amount present in cell-free samples. The MagMAX Cell-Free DNA Isolation Kit addresses this with its magnetic bead-based purification format which allows you to easily process a wide range of sample input volumes, from 100 μL to 10 mL. When used with either a KingFisher™ Duo Prime or KingFisher™ Flex magnetic particle processor, 6 to 24 plasma/serum samples of 2 mL input volume can be isolated at once. Alternatively, samples can be processed manually with the use of a magnetic stand.

Isolating and analyzing circulating cfDNA from plasma, serum, or urine has the potential to serve as a non-invasive approach to detect and monitor targets associated with certain disease states such as cancer. The first step in attaining this valuable information is efficient nucleic acid isolation that specifically recovers the circulating cfDNA while leaving the larger DNA molecules behind. This helps ensure that the shorter, fragmented DNA is concentrated and ready for downstream analysis using real-time PCR, digital PCR, or next-generation sequencing.

Benefits of MagMAX magnetic bead–based purification
Magnetic beads offer many benefits compared to other technologies for isolating cfDNA. Beads bind the DNA more efficiently than glass fiber filters, resulting in higher and more consistent yields. Additionally, because filters and vacuum manifolds are not used, there is no risk of cellular particulates clogging these items during the extraction process. This clogging issue is of particular concern with protein-rich, large-volume samples such as plasma that are commonly used in cfDNA studies.

Optimized surface chemistry
The Dynabeads MyOne Silane contained in the MagMAX Cell-Free DNA Isolation Kit have an optimized silica-like surface chemistry with high specific surface area that allows efficient kinetics and high sensitivity in nucleic acid capture. These aspects make Dynabeads MyOne Silane ideal for the recovery of circulating cfDNA targets present at low concentrations in cell-free body fluids. Once captured, these rare DNA fragments can then be eluted in small elution volumes of 15 μL to 50 μL and are ready for downstream applications.

The number of samples that can be isolated with MagMax Cell-Free DNA Isolation Kit (Cat. No. A29319) depends on the sample volume. The MagMAX Cell-Free DNA Isolation Kit provides a wide sample volume input range from 500 µL to 10 mL of plasma/serum or urine. For 2 mL samples, approximately 50 isolations can be carried out.

Can the MagMAX Cell-Free DNA Magnetic Beads within the MagMAX Cell-Free DNA Isolation Kit (Cat. No. A29319) be stored at room temperature?

If unopened, the MagMAX Cell-Free DNA Magnetic Beads within the MagMAX Cell-Free DNA Isolation Kit will be stable at room temperature for several weeks. However, for optimal performance, we recommend storing them 2-8 degrees C.

We are getting a lot of variation in cell-free DNA yield from donor to donor when using the MagMAX Cell-Free DNA Isolation Kit. Do you have any tips for how to improve that?

Levels of cell-free DNA in circulation can range from 1 ng/mL to 100 ng/mL of plasma or serum depending on the donor. For samples containing low levels of cell-free DNA, we recommend increasing the starting sample volume.

There is an abundance of gDNA in the eluate I obtained from the MagMAX Cell-Free DNA Isolation Kit (Cat. No. A29319). Do you have any recommendations to prevent that from happening?

Yields from hemolytic plasma, lipemic plasma, or other compromised samples types vary greatly from donor to donor. We recommend processing these types of samples using the manual protocol.

Can I edit the manufactured KingFisher Flex protocols?

No, manufactured protocols are locked for editing, but they can be renamed using the ‘save as' function and edited as a new version.

Can I use magnetic beads from other vendors with the KingFisher Flex instrument?

For nucleic acid extraction, we recommend using our MagMAX kits and MagJET kits. Other magnetic beads should work well as long as they are 0.5 to 10 µm in size. However, you will have to write your own KingFisher protocols using the Thermo Scientific BindIt software and validate/optimize the protocol on your own.

I am working with liquid samples with high volume and want to extract nucleic acid from them using the KingFisher Flex instrument. Can I use more volume than the protocol advises?

Yes, this can be done by sample splitting. Set the protocol so that the magnet will collect samples from multiple wells before continuing to the next step. Be sure not to overload.

Can I scale up my reaction volume with the KingFisher Flex instrument?

Yes, reaction volumes can be scaled up, and all other volumes (i.e. wash volumes) will have to be increased by the same fold difference. It is generally not recommended to scale up reaction volumes more than 2-fold unless the plate's volume capacity will allow it.
