The average particle size for the Pierce Streptavidin Agarose beads is 45 to 165 µm.
The antigen:antibody ratio will need to be determined empirically since one cannot be sure of the amount of specific antibody against the antigen present in the plasma. The starting point is the binding capacity of the High Capacity Streptavidin Agarose Resin (Cat. No. 20347) which is 1-3 mg of biotinylated BSA/mL of settled resin. The binding capacity will vary depending on the molecular weight of the biotinylated antigen. Let's assume the molecular weight of the antigen is close to that of BSA (66.4 kDa), you can use an average of 2 mg antibody or antigen/mL of resin to do the calculation. Determine how much resin they want to use and calculate how much biotinylated protein is needed and then empirically determine how much plasma is needed.

Streptavidin is a protein composed of four identical subunits, each containing a high affinity binding site for biotin (K-D = 10 -15 M) . Streptavidin has the same biotin binding properties as avidin, but it has a low isoelectric point (pI=5) and no carbohydrate groups, resulting in low non-specific binding.
