Applied Biosystems™ MagMAX™ HMW DNA Kit and Booster
Description
The MagMAX HMW DNA Kit is a magnetic bead-based nucleic acid purification kit for isolation of high-quality, high molecular weight (HMW) DNA from whole blood, cells, and tissue. This versatile kit is optimized for both automated workflows with KingFisher purification systems and manual isolation processes, providing flexibility and scalability. It is specifically designed to obtain intact DNA fragments larger than 100 kb, making it excellent for long-read sequencing and various downstream molecular applications.
The MagMAX HMW DNA Kit is for use with fresh and frozen whole blood, while the MagMAX HMW DNA Kit with Cell and Tissue Extraction Buffer is also compatible with cell and tissue sample types. Already have MagMAX DNA Multi-Sample Ultra 2.0 and/or MagMAX Cell and Tissue Extraction Buffer? No problem, we offer MagMAX HMW DNA Booster separately in the 100 prep size found in the kit or a 1000 prep size for higher throughput.
Features of the MagMAX HMW DNA Kit include:
- Reproducibility and efficiency—consistently yields intact DNA fragments exceeding 100 kb from a range of sample types
- Versatile solution—flexible workflows for low- to high-throughput sample processing with manual or automated protocols
- Fast and optimized workflow—eliminates the need for post-processing steps or overnight elution
- Innovate your research—optimal performance with long-read systems such as Oxford Nanopore Technologies and PacBio systems
- Automation compatibility
- Available as stand-alone products—reagents available in larger volume bottles sold separately to meet your scalability needs
Versatility for downstream applications
Start your workflow out right with the MagMAX HMW DNA Kit, designed to meet the high demand for high-quality nucleic acid isolation, a vital component for highly sensitive downstream applications. The MagMAX HMW DNA Kit has optimized workflows available with the KingFisher purification systems, enabling a semi-automated workflow with a total turnaround time of less than two hours. This versatile kit is also optimized for lower throughput manual workflows, making it an excellent choice for laboratories of all sizes and throughput needs. Experience the perfect blend of efficiency, flexibility, and reliability with the MagMAX HMW DNA Kit for your downstream molecular applications, such as long-read sequencing.
Isolate HMW DNA from multiple sample types with a single sample prep kit
The MagMAX HMW DNA Kit leverages the innovative MagMAX HMW DNA Booster to enhance the elongation of fragment size. It is designed to isolate large fragment gDNA (100–300 kb) from various sample types, including 200 μL of fresh or frozen whole blood, one million cultured cells, and 2–8 mg tissue samples. Dynabeads magnetic beads with optimized surface chemistry and high specific surface area enable efficient kinetics and high sensitivity in capturing nucleic acids. The robust chemistry and workflow design minimizes impurities, such as proteins and detergents, that can interfere with downstream molecular analysis. Finally, the HMW genomic DNA is released from the beads using a low quantity TE elution solution.
The MagMAX HMW DNA Kit leverages the MagMAX DNA Multi-sample Ultra 2.0 Kit chemistry. This kit is specifically designed for the extraction of high molecular weight DNA from fresh and frozen whole blood samples. For cell and tissue samples, the MagMAX HMW DNA Kit with Cell and Tissue DNA Extraction Buffer provides an optimized solution, delivering reliable and efficient nucleic acid isolation. Existing users of the MagMAX DNA Multi-sample Ultra 2.0 Kit (A36570) can enhance their workflow by incorporating the MagMAX HMW DNA Booster (A36624, A36625) for whole blood, along with the MagMAX Cell and Tissue DNA Extraction Buffer (A45469, A45470) for cells and tissue. This combination enables the isolation of longer DNA fragments, thus broadening the scope of your genomic research.

The isolated DNA recovered would likely not achieve greater than 100 kb in fragment size and the workflow reproducibility of the HMW kit would be compromised. If the customer wishes to leave the Booster Solution out of the workflow, we recommend following the guidance of Applied Biosystems(TM) MagMAX(TM) DNA Multi-Sample Ultra 2.0 kit to achieve genomic DNA without expanding into the HMW (>100 bp) or Ultra-HMW DNA (around 300 bp) recovery range.
Fragile samples can get damaged when they are forced through the narrow orifice of standard pipette tips. Using a combination of standard and wide bore pipette tips helps to increase the yield of HMW DNA, keeping the fragment sizes intact. Reference the MagMAX HMW DNA User Guide (MAN1001478).
Yes, low-bind tubes are recommended to minimize loss due to absorption to the tube walls. Please refer to MagMAX HMW DNA User Guide (MAN1001478) for guidance on when to transfer to new tubes to reduce risk to sample contamination.
The enhancer solution improves proteinase K activity at optimal concentrations; however, excessive enhancer solution can reduce proteinase K effectiveness. For blood and cell samples, adding the blood sample and cell lysate in the correct order dilutes the enhancer to achieve optimal proteinase K activity during the digestion step. For tissue samples, the tissue must be digested with proteinase K before using the tissue lysate for extraction. Preparing a master mix with Cell/Tissue extraction buffer ensures the enhancer solution is diluted to the concentration needed for optimal proteinase K activity in these samples.
