The MagMAX DNA Multi-Sample Ultra 2.0 Kit is an automatable magnetic bead-based sample preparation technology for the isolation of genomic DNA (gDNA) from 50 μL to 2 mL of whole blood, saliva, buffy coat, and buccal swabs. The resulting purified gDNA is ideal for a variety of downstream molecular biology applications such as qPCR, next-generation sequencing, and microarray analysis. One kit is sufficient to process 200 50–200 μL samples or 100 200–400 μL samples except for buffy coat samples. For buffy coat only, one kit is sufficient to process 40–60 samples at 50–200 uL input volume. To process up to 200 buffy coat samples, please purchase additional Elution Solution (Cat. No. A36582).

Isolate genomic DNA from multiple sample types with a single sample prep kit
The MagMAX DNA Multi-Sample Ultra 2.0 Kit isolates gDNA from 50 μL–2 mL of whole blood (figure below), fresh and stabilized saliva (figure below), and EDTA/Streck cfDNA tube buffy coat (figure below) samples. Additionally, the same kit can be used to isolate gDNA from buccal swabs and other biological sample types. The MagMAX DNA Multi-Sample Ultra 2.0 Kit maximizes gDNA yields by treating samples with proteinase K (PK) and PK-enhancer followed by an innovative lysis/binding solution to simultaneously lyse cells, denature proteins, and capture gDNA on the high-binding capacity Ultra 2.0 gDNA magnetic binding beads. The Ultra 2.0 Wash I solution removes proteins, detergents, and other impurities from the bound nucleic acid. The gDNA is then released from the beads using a low-TE elution solution.

The MagMAX DNA Multi-Sample Ultra 2.0 Kit is optimized for use on the moderate-throughput KingFisher Duo Prime (Cat. No. 5400110) and the higher-throughput KingFisher Flex (Cat. No. 5400630) purification systems. Users can process up to 400 μL of sample volume in high-throughput SBS 96-well deep-well microtiter plates using the Flex system. For smaller sample batches, users can process 12 samples using the Duo Prime system. Larger sample input volumes (1–2 mL), which are more challenging to process, are purified in SBS 24-well deep-well microtiter plates for either 24 samples on the Flex or 6 samples on the Duo Prime system. KingFisher instrument BindIt software scripts for all sample types and both Flex and Duo Prime systems can be downloaded below and installed onto the KingFisher instruments.

We would recommend using one of our MagMAX DNA Multi-Sample Ultra 2.0 Kit or ChargeSwitch gDNA Buccal Cell Kits, which were designed to isolate high-quality genomic DNA from buccal samples. We also offer normalized kits, which will normalize isolated DNA concentration from sample to sample.

What will happen if the MagMAX HMW DNA Booster is not added during the workflow?

The isolated DNA recovered would likely not achieve greater than 100 kb in fragment size and the workflow reproducibility of the HMW kit would be compromised. If the customer wishes to leave the Booster Solution out of the workflow, we recommend following the guidance of Applied Biosystems(TM) MagMAX(TM) DNA Multi-Sample Ultra 2.0 kit to achieve genomic DNA without expanding into the HMW (>100 bp) or Ultra-HMW DNA (around 300 bp) recovery range.
