AccuPrime™ Taq DNA Polymerase High Fidelity (40μL, at 5U/μL)) is supplied with: 50mM MgSO4 (1mL), 10X AccuPrime™ PCR Buffer I (1mL), 10X AccuPrime™ PCR Buffer II (1mL)

Buffer I and II differ in their concentration of thermostable AccuPrime™ protein. Their components are: 600mM Tris-SO4 (pH 8.9), 180mM (NH4)2SO4, 20mM MgSO4, 2mM dGTP, 2mM dATP, 2mM dTTP, 2mM dCTP, thermostable AccuPrime™ protein, 10% glycerol. The supplied PCR Buffer is a 10X concentrate and should be diluted for use. Buffer I is optimized for plasmids, cDNA, and λDNA. Buffer II is optimized for genomic DNA.

It includes an enzyme blend of Platinum Taq DNA Polymerase and a proofreading enzyme, plus AccuPrime accessory proteins for improved PCR fidelity, yield, and specificity over other hot-start DNA polymerases.

How it works

Pyrococcus species GB-D polymerase is a proofreading enzyme that possesses a 3' to 5' exonuclease activity. Mixing this enzyme with Taq DNA Polymerase increases fidelity and allows amplification of simple and complex DNA templates over a large range of target sizes.

The Platinum antibody complexes with Taq DNA polymerase and inhibits activity at room temperature. Activity is restored after the initial denaturation step, providing an automatic hot start PCR.

The thermostable AccuPrime accessory proteins enhance specific primer-template hybridization during every cycle of PCR, preventing mispriming and enhancing PCR specificity and yield.
