Tempus Blood RNA Tubes are for stabilization and isolation of total RNA from whole blood for gene expression analysis.

Tempus Blood RNA Tubes are for stabilization and isolation of total RNA from whole blood for gene expression analysis. Advantages of Tempus Blood RNA Tubes:
• Immediately lyse whole blood cells and stabilize RNA in a single step—no RBC lysis or proteinase K treatments required
• Freeze global gene expression profiles for up to five days at room temperature; greater than seven days at 4°C

How it works
Each Tempus Blood RNA Tube contains 6 mL Stabilizing Reagent. When the blood (3 mL) is drawn into the tube and mixed with this reagent, lysis occurs almost immediately. The stabilizing reagent inactivates cellular RNases and selectively precipitates RNA; genomic DNA (gDNA) while proteins remain in solution. Blood drawn into the Tempus Blood RNA Tube is stable for up to five days at room temperature, or minimally seven days at 4°C.

Accessory product
When the Tempus Blood RNA Tubes are used with the Tempus Spin RNA Isolation Kit (Cat. No. 4380204), RNAs with greater stability and higher yields of RNA are produced.

I am doing blood work and using the Tempus collection tubes. Is the Tempus system compatible with the RiboPure-Blood RNA Purification Kit?

No, please follow the RNA isolation methods described with the Tempus system. The collection tubes are not compatible with the RiboPure kit.

What is the recommended storage temperature and stability of samples in the Tempus Blood RNA Tube?

Our general recommendations are that once blood has been collected in the Tempus Blood RNA Tubes, you can store the tube at room temperature for up to 5 days or at 4 degrees C for up to 7 days. For long term storage, the tube can be frozen at -80 degrees C for at least 1.5 years.

Do you offer the Stabilizing Reagent that comes inside the Tempus Blood RNA Tube (Cat. No. 4342792) as a stand-alone item?

We do not offer the Stabilizing Reagent that comes inside the Tempus Blood RNA Tube as a stand-alone item.

Can animal blood stored in Tempus Blood RNA Tubes or PAXgene tubes be prepared with the MagMAX for Stabilized Blood Tubes RNA Isolation Kits (Cat. Nos. 4451893 & 4451894)?

Equine whole blood has been successfully tested using this system, getting RINs around 9. Bovine blood should work similarly since there is not much of a difference between the two species for RNA isolation from whole blood, but this has not been tested. The protocol would be the same as the user guide and samples should be mixed vigorously immediately after blood is drawn into the tube for at least 10 secs, incorporating tube inversion to be sure the sample is stabilized.

Will the RNA stabilization solution in the Tempus Blood RNA Tube (Cat. No. 4342792) lyse E. coli cells that are in the blood samples?

The RNA stabilization solution in Tempus Blood RNA Tubes will not lyse E. coli cells at ambient temperature. If lysis of gram-negative bacteria is desired, mix the sample thoroughly with the RNA stabilization solution, and heat the lysate at 95 degrees C for 5 mins.

What is the DNA yield from 3 mL of blood that is stored in a Tempus Blood RNA Tube (Cat. No. 4342792)?

The yield for 500 µL of the RNA-depleted Tempus lysate, which is equivalent to 125 µL of whole blood, ranges from 2-5 µg

Can I isolate DNA from blood stored in a Tempus Blood RNA Tube (Cat. No. 4342792)?

DNA can be isolated from the supernatant of blood samples stored in Tempus Blood RNA Tubes once the RNA is pelleted.

What level of yield and quality should I expect for RNA extracted with Tempus Blood RNA Tubes and PAXgene Blood RNA Tubes?

For Tempus/PAXgene Blood RNA Tubes, here is the expected RNA yield and quality:
Yield: 3-25 µg
Quality (RIN): ≥7
Quality (A260/A280): 1.8-2.2 gDNA (w/w): <1.56%

Do Tempus Blood RNA Tubes (Cat. No. 4342792) stabilize RNA similarly to RNAlater Stabilization Solution (Cat. No. AM7021)?

Both Tempus Blood RNA Tubes and RNAlater Stabilization Solution employ the same effective stabilization of mRNA expression profiles, and eliminate the need to isolate the RNA immediately after sample collection. However, there are some key differences:
- Components in each stabilization solution are different. The exact components and differences are proprietary.
- The RNAlater Stabilization Solution preserves RNA from degradation without lysing blood cells.
- Samples in RNAlater Stabilization Solution can be safely stored for up to 1 week at 25 degrees C, or up to 1 month at 4 degrees C.
