ELISA

The Human TECK/CCL25 ELISA quantitates Hu TECK in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu TECK. Principle of the method The Human TECK solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Rigorous validation Each manufactured lot of this ELISA kit is quality tested for criteria such as sensitivity, specificity, precision, and lot-to-lot consistency. See manual for more information on validation.

Chemokines are likely to play an important role in regulating the trafficking of developing T cells within the thymus. Chemokine C-C thymus expressed chemokine (TECK), also designated chemokine ligand 25 (CCL25), small induc-ible cytokine A25, chemokine b-15 or CK b-15, is expressed predominantly in thymic dendritic cells, thymic epithelial cells and in the small intestine. TECK, a CCR9 ligand, has suppressive activity against immature subsets of myeloid progenitors which have been stimulated to proliferate by multiple growth factors. TECK delivers signals through CCR9, which is important for the nav-igation of developing thymocytes. Bone marrow pre-pro-B cells and cells capable of generating pro-B colonies in the presence of interleukin-7 and Flt-3 ligand migrate to TECK, a response lost in later stages of B cell development.
