Thermo Scientific™ Syn-PER Synaptic Protein Extraction Reagent is a proprietary cell lysis reagent for efficient isolation of synaptosomes containing functional synaptic proteins from neuronal tissue and primary cultured neurons.

We can not guarantee that the total synaptic yield from cultured neurons should be comparable to using fresh tissue.
We have tested using Syn-PER Synaptic Protein Extraction Reagent with frozen (mouse) brain tissue and total synaptic protein yields were comparable to fresh brain tissue.
Please note, frozen tissues are not recommended for synaptosome function tests such as neurotransmitter release and up take assays because synaptosomes from frozen tissues are not viable. This is a difference between frozen and fresh tissue in terms of application.
Yes. RNA extraction can be performed after using Syn-PER Synaptic Protein Extraction Reagent (Cat. No. 87793).
After the lysis step, you will need to perform two centrifugations (step 4 and 5 in the manual, the first at low speed to discard the tissue or cell debris. The second one, at higher speed, will pellet synaptosomes. The supernatant arising from this step will include the cytosolic fraction (step 6).
If the synaptosomes are intact and have RNA inside, the RNA should be extractable by the kit or method of your choice.
Please note: This extraction procedure should be experimentally tested.
