Invitrogen™ Human RAF1 (cRAF) [Y340D, Y341D], GST Tag Recombinant Protein
Description
For maximum recovery please spin prior to use. Unless noted below, aliquots of the 5 μg, 10 μg and 20 μg sizes of kinase are not recommended as materials can be used in original packaging until exhausted. For larger sizes, the number of freeze/thaws may be reduced by preparing aliquots, aliquots below 20 μL are not recommended. Please never store a kinase diluted. If properly stored at -80C, this product is guaranteed for 6 months from date of purchase. Protein Form: Recombinant, Catalytic, Mutant.
RAF1 (cRAF) is a MAP kinase kinase (MAP3K) which functions downstream of the Ras family of membrane associated GTPases to which it binds directly. Once activated RAF1 can phosphorylate and activate the dual specificity protein kinases MEK1 and MEK2 which in turn phosphorylates and activates the serine/threonine specific protein kinases ERK1 and ERK2. The RAF1 Y340D Y341D mutant is constitutively active. RAF1 functions downstream of the Ras family of membrane associated GTPases to which it binds directly. Once activated Raf-1 can phosphorylate to activate the dual specificity protein kinases MEK1 and MEK2 which in turn phosphorylate to activate the serine/threonine specific protein kinases ERK1 and ERK2. Activated ERKs are pleiotropic effectors of cell physiology and play an important role in the control of gene expression involved in the cell division cycle, apoptosis, cell differentiation and cell migration. Mutations in this gene are associated with Noonan syndrome 5 and LEOPARD syndrome 2.
