ELISA

The Human Platelet Derived Growth Factor-Receptor Beta (Hu PDGFRB) ELISA quantitates Hu PDGFRB in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu PDGFRB. Principle of the method The Human PDGFRB solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. PDGFRB is a receptor tyrosine kinase and has been implicated in atherogenesis and in cell transformation by the SIS oncogene. These growth factors are mitogens for cells of mesenchymal origin. The identity of the growth factor bound to a receptor monomer determines whether the functional receptor is a homodimer or a heterodimer, composed of both platelet-derived growth factor receptor alpha and beta polypeptides. The gene is flanked on chromosome 5 by the genes for granulocyte-macrophage colony-stimulating factor and macrophage-colony stimulating factor receptor; all three genes may be implicated in the 5-q syndrome. A translocation between chromosomes 5 and 12, that fuses PDGFRB (CD140b) to that of the translocation, ETV6, leukemia gene, results in chronic myeloproliferative disorder with eosinophilia.
