MPP5 (Membrane Protein, Palmitoylated 5) plays a critical role in neurogenesis and hematopoiesis. This protein is crucial for establishing and maintaining cell polarity in epithelial cells and neurons. It is involved in the Par-3 complex, which contributes to maintaining tight junctions in epithelial cells. Mutations in MPP5 have been associated with severe neurologic impairments, including global developmental delay (GDD) and behavioral changes. In animal models, MPP5 is highly involved in the early differentiation of multipotent progenitors and supports long-term lineage-specific contributions, primarily in myeloid and lymphoid cells.
