Insulin-Transferrin-Selenium (ITS-G) Select Supplement is an animal-origin-free (AOF) basal medium supplement that is used to reduce the amount of FBS needed to culture cells.

Gibco Insulin-Transferrin-Selenium (ITS-G) Select Supplement is an animal origin-free (AOF) basal medium supplement. The insulin and transferrin in the ITS-G Select formulation are produced in non-mammalian recombinant expression systems.

ITS-G is used as a basal medium supplement to reduce the amount of fetal bovine serum (FBS) needed to culture cells. Originally used with RPMI 1640 and Minimum Essential Medium (MEM), ITS-G is now routinely used with other basal media to support a wide variety of cell types in the presence of 2–4% FBS.

Insulin promotes glucose and amino acid uptake, lipogenesis, intracellular transport, and the synthesis of proteins and nucleic acids. Transferrin is an iron carrier and may also help reduce toxic levels of oxygen radicals and peroxide. Selenium, as sodium selenite, is a co-factor for glutathione peroxidase and other proteins and is used as an antioxidant in media.

The product formulation contains the following components:

  • Insulin (1000.0 mg/L)
  • Transferrin (550.0 mg/L)
  • Sodium selenite (0.67 mg/L)

cGMP manufacturing and quality system

ITS-G Select Supplement is manufactured at a cGMP-compliant facility located in Grand Island, New York. The facility is registered with the FDA as a medical device manufacturer and is certified to ISO 13485 standards.

What is the buffer composition of the various Insulin-Transferrin-Selenium (ITS) (100X) and Insulin-Transferrin-Selenium (ITS) Select (100X) supplements?

All ITS and ITS Select supplements are formulated in Earle's Balanced Salt Solution (EBSS).

What is the difference between Insulin-Transferrin-Selenium (ITS) (100X) and Insulin-Transferrin-Selenium (ITS) Select (100X) supplements?

ITS Select supplements are animal origin-free (AOF) versions of the ITS supplements offered by Thermo Fisher Scientific.

ITS Select supplements use recombinant versions of both proteins in the formulation: insulin and transferrin.

All other versions of ITS, those without the Select designation, utilize recombinant insulin and a human-derived holo transferrin.

What are the individual concentrations of insulin, transferrin, and selenium in the Insulin-Transferrin-Selenium (ITS) (100X) supplements?

All ITS and ITS Select supplements have the following concentrations of their components:

Insulin: 1.0 g/L
Transferrin: 0.55 g/L
Selenium: 0.00067 g/L

Can I store aliquots of Insulin-Transferrin-Selenium (ITS) (100X) supplements at -20 degrees C without compromising the integrity of the product?

We recommend storing all ITS and ITS Select supplements at 2-8 degrees C as there is a chance that the insulin will precipitate upon freezing and thawing.

Are all Insulin-Transferrin-Selenium (ITS) (100X) supplements offered by Thermo Fisher Scientific animal origin-free?

We offer both animal origin and animal origin-free versions of our ITS supplements. The animal origin-free product versions are referred to as ITS Select supplements.

Insulin-Transferrin-Selenium (ITS-G) (100X) (Cat. No. 41400045), Insulin-Transferrin-Selenium-Ethanolamine (ITS-X) (100X) (Cat. No. 51500056), and Insulin-Transferrin-Selenium-Sodium Pyruvate (ITS-A) (100X) (Cat. No. 51300044) are not animal origin-free.

Animal origin-free product versions are available for the ITS-G and ITS-X formulations: Insulin-Transferrin-Selenium (ITS-G) Select Supplement (100X) (Cat. No. A4000046501 and A4000046502) and Insulin-Transferrin-Selenium-Ethanolamine (ITS-X) Select Supplement (100X) (Cat. No. A4000046401 and Cat. No. A4000046402.

Can I store Insulin-Transferrin-Selenium (ITS) (100X) supplements at -20 degrees C?

All ITS and ITS Select supplements have a shelf life of between 12 to 18 months from date of manufacture when stored at 2-8 degrees C (see product pages for specific shelf-life). In general, we do not recommend storing these products at -20 degrees C due to the potential issue of proteins precipitating from the solution upon thawing.

What is the source of transferrin in the Insulin-Transferrin-Selenium (ITS) supplements? Is it APO or HOLO?

Our ITS supplements contain human-derived holo transferrin and our ITS Select supplements contain a partially saturated recombinant human transferrin produced in a plant-based expression system, that performs similarly to human-derived holo transferrin in cell culture applications. This information can also be accessed in the Certificate of Origin (COO).

The following ITS supplements use human-derived holo transferrin: Insulin-Transferrin-Selenium (ITS-G) (100X) (Cat. No. 41400045), Insulin-Transferrin-Selenium-Ethanolamine (ITS-X) (100X) (Cat. No. 51500056), and Insulin-Transferrin-Selenium-Sodium Pyruvate (ITS-A) (100X) (Cat. No. 51300044).

The following ITS Select supplements use recombinant human transferrin produced in a plant-based expression system: Insulin-Transferrin-Selenium (ITS-G) Select Supplement (100X) (Cat. No. A4000046501 and A4000046502) and Insulin-Transferrin-Selenium-Ethanolamine (ITS-X) Select Supplement (Cat. No. A4000046401 and Cat. No. A4000046402).

What is the source of insulin in the Insulin-Transferrin-Selenium (ITS) supplements?

Recombinant human insulin is used in all ITS and ITS Select supplements.

What concentration of Insulin-Transferrin-Selenium (ITS) supplements should I use?

All ITS and ITS Select supplements are provided at 100X concentration. These can be added to media at 10mL per 1L of media. This can reduce the amount of FBS required (typically to concentrations of 2- 4%).

What is the advantage of using Insulin-Transferrin-Selenium (ITS-G) supplements in cultures?

ITS-G and ITS-G Select supplements have been designed to supplement basal medium to support the growth of various cell types at reduced FBS concentrations.

What is the advantage of using Insulin-Transferrin-Selenium (ITS) supplements in cultures?

ITS and ITS Select supplements are used to reduce the amount of fetal bovine serum (FBS) required for routine maintenance and low-density attachment of many cell types. They may also be used for supplementation of lab-developed media formulations.

What do Insulin-Transferrin-Selenium (ITS) supplements contain?

ITS and ITS Select supplements contain insulin, transferrin, and sodium selenite. In addition to these components, ITS-X and ITS-X Select supplements contain ethanolamine and ITS-A supplement contains sodium pyruvate.
