Interleukin-1 receptor type 1 (UniProt P14778; also known as CD121 antigen-like family member A, CD121a, IL-1R-1, IL-1R-alpha, IL-1R1, IL-1RT-1, IL-1RT1, Interleukin-1 receptor alpha, Interleukin-1 receptor type I, p80) is encoded by the IL1R1 (also known as IL1R, IL1RA, IL1RT1) gene (Gene ID 3554) in human. The biological effects of interleukin-1 (IL-1 and IL-1 ), such as acute and chronic inflammation, are mediated by the membrane-bound IL-1 type 1 receptor (IL-1R1). IL-1 binding to the extracellular domain of IL1R1 promotes the recruitment of the IL-1 receptor accessory protein and subsequent signal transduction mediated by the cytoplasmic domains of IL-1R1 and IL1-R2. The IL-1 type 2 receptor (IL1R2) is unable to initiate signaling and only acts as a decoy receptor. Soluble receptors for IL-1 (sIL-1Rs) are produced by proteolytic cleavage of membrane-bound receptors or by alternative splicing and act as natural inhibitors by competing against membrane-bound IL1Rs for IL-1 binding. IL-1R1 is predominantly expressed on endothelial cells, smooth muscle cells, epithelial cells, hepatocytes, fibroblasts, keratinocytes, epidermal dendritic cells and T lymphocytes, while IL-1R2 is expressed on monocytes, neutrophils, T and B lymphocytes. Human IL-1R1 is produced with a signal peptide sequence (a.a. 1-17) that is cleaved posttranslationally to yield the mature receptor with an N-terminal extracellular region (a.a. 18-336) composed of three C2-type Ig-like domains (a.a. 23-110, 118-210, 226-328), followed by a transmembrane domain (a.a. 337-356) and a cytoplasmic tail (a.a. 357-569) that contains the Toll/interleukin-1 receptor (TIR) homology domain (383-541).
