Description
Carrier-Free Reconstitution: We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Reconstitute lyophilized recombinant human Thrombopoietin in PBS containing 0.1% human serum albumin. Further dilutions should be made in low endotoxin medium or a buffered solution containing a carrier protein such as heat inactivated FCS or tissue culture grade BSA. Storage: Store this lyophilized preparation at <-20°C, preferably desiccated. Upon reconstitution, apportion into working aliquots and store at <-20°C. Avoid repeated freeze/thaw cycles.
Thrombopoietin (TPO, THPO, MPL ligand, megakaryocyte colony-stimulating factor) is responsible for the generation of platelets through its action on hematopoietic cells committed to the megakaryocyte lineage. Binding of thrombopoietin to its receptor, MPL (CD110), initiates a signaling cascade via JAK/STAT, MAPK, and PI3K/AKT signaling pathways that leads to the proliferation and differentiation of megakaryocytes and, ultimately, to the production of platelets (thrombopoiesis). Thrombopoietin is constitutively produced by the liver and has a predicted molecular weight of 38 kDa.
