ELISA

The Human Cortisol ELISA quantitates Cortisol in serum, plasma, urine, saliva. Principle of the method The Human Cortisol solid phase competitive ELISA (enzyme linked immunosorbent assay) is designed to measure the amount of the target bound between the pre coated antigen and the HRP linked antibody specific for Human Cortisol. A target specific antigen has been pre coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells, where Human Cortisol in the samples (or standards) competes with the immobilized (capture) antigen for sites on the HRP linked detection antibody. Excess conjugate and unbound sample or standard are washed away from the plate. The substrate solution is added to each well, and the enzyme substrate reaction is terminated by the addition of a stop solution, turning the color yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450±2 nm. The intensity of this signal is inversely proportional to the concentration of Human Cortisol present in the original specimen, which is determined by comparing the OD of the samples to the standard curve. Cortisol is the most potent glucocorticoid produced by the human adrenal. It is synthesized from cholesterol and its production is stimulated by pituitary adrenocorticotropic hormone (ACTH) which is regulated by corticotropin releasing factor (CRF). ACTH and CRF secretions are inhibited by high cortisol levels in a negative feedback loop. In plasma a majority of cortisol is bound with high affinity to corticosteroid binding globulin (CBG or transcotin). Cortisol acts through specific intracellular receptors and affects numerous physiologic systems including immune function, glucose counter regulation, vascular tone, and bone metabolism.
