ELISA

The Human Bone Morphogenic Protein 7 (Hu BMP-7) ELISA quantitates Hu BMP-7 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu BMP-7. Principle of the method The Human BMP-7 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. The bone morphogenetic proteins (BMPs) are a family of secreted signaling molecules that can induce ectopic bone growth. Many BMPs are part of the transforming growth factor-beta (TGFB) superfamily. BMPs were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extra skeletal site. Based on its expression early in embryogenesis, BMP7 has a proposed role in early development. In addition, the fact that BMP7 is closely related to BMP5 and BMP6 has lead to speculation of possible bone inductive activity.
