Degrade unwanted bacterial cell wall in protein or nucleic acid extracts

Lysozyme is an enzyme used to break down bacterial cell walls to improve protein or nucleic acid extraction efficiency. Lysozymes (muramidases) are a family of enzymes with antimicrobial activity characterized by the ability to damage the cell wall of bacteria. The enzyme acts by catalyzing the hydrolysis of 1,4-beta-linkages between N-acetylmuramic acid and N-acetyl-D-glucosamine residues in peptidoglycans and between the N-acetyl-D-glucosamine residues in chitodextrins. Although hen egg white lysozyme is most effective for the lysis of gram-positive bacteria, it also facilitates the lysis of gram-negative bacteria such as Salmonella and Shigella. The lysis of E. coli is especially improved by the addition of both lysozyme and a nucleases such as DNase I.

How should I prepare and store Lysozyme (Cat. No. 89833)?

Lysozyme powder can be dissolved in 0.1 M KH2PO4 (pH 6.24) to the desired concentration - up to 5 mg/mL. Once the solution has been prepared, it can be stored at 4 degrees C for up to 5-6 weeks, or at room temperature for up to 6-7 days, without affecting performance.

Can you explain how the B-PER Bacterial Protein Extraction Reagent lyses cells?

The B-PER Reagent solution contains a proprietary, mild, non-ionic detergent in 20 mM Tris-HCl, pH 7.5. It effectively disrupts cells and solubilizes native or recombinant proteins without denaturation. The reagent creates holes in the cell membrane that will leak out cytosolic proteins. The sample may become very viscous when the bacterial chromosome is released. We recommend adding DNAse I (Cat. No. 90083) to the reagent to reduce viscosity. For better lysis efficiency and if there are inclusion bodies, we recommend adding Lysozyme (Cat. No. 90082) to the reagent. Alternatively, you may purchase the B-PER Bacterial Protein Extraction Reagent with Enzymes Kit (Cat. No. 90078 or 90079) that includes the B-PER Bacterial Protein Extraction Reagent, DNase I, and Lysozyme.
