For use in the agarose overlay when performing the plaque assay

Grace's Supplemented Insect Medium was originally designed for the growth of cells from the Australian Emperor Gum Moth, Antherea eucalypti. Used for the growth of Spodeptera frugiperda cells, Sf9 and Sf21. Sf9 and Sf21 cells are routinely used to express recombinant protein using the Baculovirus Expression Vector System (BEVS). Modified as follows: With Lactalbumin hydrolysate, Yeastolate, L-glutamine and Sodium bicarbonate. The complete formulation is available. Grace's Supplemented Insect Medium, 2x contains lactalbumin hydrolysate and yeastolate not commonly found in the unsupplemented version commonly diluted with sterile water, Fetal bovine Serum and agarose before use in the agarose overlay, as described in the product manual.

What additional supplements are required in Grace's Insect Cell Culture Medium, Unsupplemented to make a "complete" formulation that will support insect cell growth?

Grace's Insect Cell Culture Medium, Unsupplemented requires addition of 3.33 g/L of lactalbumin hydrolysate and 3.33 g/L of yeastolate for the “base” growth medium. For the “complete” growth medium that will support insect cell growth, 10% fetal bovine serum must also be added. This “complete” growth medium is also referred to as TNM-FH.

What medium do you recommend to grow Sf9 or Sf21 cells for performing plaque assays?

We would recommend using our Grace's Supplemented Insect Medium (Cat. No. 11667037) or SF-900 Medium (1.3X) (Cat. No. 10967032). Grace's Supplmented Insect Media is a 2X formulation intended for use in the agarose overlay when performing the plaque assay. The media has been modified with:
- Lactalbumin hydrolysate
- Yeastolate
- L-glutamine
- Sodium bicarbonate
