CTS™ OpTmizer™ T Cell Expansion SFM, bottle format

GIBCO OpTmizer™ CTS™ T-Cell Expansion SFM has been developed for the growth and expansion of human T lymphocytes. OpTmizer™ CTS™ T-Cell Expansion medium is a complete serum-free, xeno-free 1X medium consisting of OpTmizer™ T-Cell Expansion Basal Medium (1 L bottle) and OpTmizer™ T-Cell Expansion Supplement (26 mL), which are mixed together prior to use.

  • Maintains similar phenotype and function (e.g., cytokine secretion profile) of polyclonally activated and cultured T cells as observed for conventional serum supplemented medium
  • Consistent performance in supporting T cell expansion
  • Supports high density T cell culture (e.g., >3x106 CD3+ T cells/mL) in static culture

    Do I need to order additional components with the CTS OpTmizer T Cell Expansion SFM (Cat. Nos. A1048501, A1048502, A1048503)?

    The CTS OpTmizer T Cell Expansion SFM (Cat. Nos. A1048501, A1048502, A1048503) kit comes with OpTmizer T Cell Expansion Basal Medium and OpTmizer T Cell Expansion Supplement, which are mixed together prior to use. Depending on your protocol you may need to purchase T cell growth factors such as IL-2 (Cat. No. PHC0023) and L-glutamine (Cat. No. 25030) to form a complete medium. To boost growth of human T cells, human AB serum can be added. If desired, antibiotics such as Gentamicin (Cat. No. 15750), can also be added.

    Will CTS OpTmizer T Cell Expansion SFM (Cat. No. A1048501, A1048502, A1048503) work with all types of T cells?

    CTS OpTmizer T Cell Expansion SFM (Cat. No. A1048501, A1048502, A1048503) has been demonstrated to be effective for the ex vivo expansion of human CD3 T cells, including polyclonal T cell populations, antigen-specific T cells, tumor-infiltrating lymphocytes (TIL), cord blood T cells, regulatory T cells, T cells from HIV-infected donors, gene-modified T cells (both lenti- and MLV-based vectors), including cells of both the CD4+ and CD8+ phenotype.

    Can CTS OpTmizer T Cell Expansion SFM (Cat. Nos. A1048501, A1048502, A1048503) be used for animal T cell cultures other than human?

    CTS OpTmizer T Cell Expansion SFM (Cat. No. A1048501, A1048502, A1048503) has only been tested on human T cells.

    Is there a difference in formulation of the CTS OpTmizer T Cell Expansion SFM between the bottle and bag packaging formats?

    The bottle and bag packaging formats of the medium have the same formulation.

    Can I use CTS OpTmizer T Cell Expansion SFM for culture and expansion of mouse T cells? If not, which medium would you recommend?

    We have not tested this medium on mouse T cells. If you're looking for a serum-free medium to culture mouse T cells, you can try AIM V Medium (Cat. No. 12055083 or 12055091). For mouse cells, we recommend adding 50 µM beta-mercaptoethanol (Cat. No. 21985023).

    Can GlutaMAX be used instead of Glutamine to supplement CTS OpTmizer T-Cell Expansion SFM?

    GlutaMAX cannot replace Glutamine because cells may not initially use GlutaMAX so that T cells cannot grow. We recommend adding 4 mM GlutaMAX and 2 mM Glutamine (as stated in the manual) into the CTS OpTmizer T-Cell Expansion SFM if GlutaMAX is needed.

    What is the shelf life of CTS OpTmizer T-Cell Expansion SFM?

    The complete CTS OpTmizer T-Cell Expansion SFM is stable for 4 weeks when stored in the dark at 2-8 degrees C. However, the two individual medium components are stable for 18 months when stored in the dark at 2-8 degrees C.
