Description

The MEGAclear Transcription Clean-up Kit is designed for rapid high-throughput purification of RNA from enzymatic reactions such as in vitro transcription. The kit efficiently separates RNA from unincorporated NTPs, enzymes, and buffer components. The process is simple and fast, taking less than 30 minutes, and it recovers from 1 ng to 500 μg of RNA efficiently. The MEGAclear kit is appropriate for purification of ssRNA larger than 100 nt and dsRNA larger than 200 bp.

The MEGAclear kit purifies transcription reactions to produce high-quality RNA, free of unincorporated NTPs, enzymes, and buffer components, ideal for any downstream application that requires purified RNA. It can be used to remove nucleotides, short oligonucleotides,proteins, and salts from RNA. The RNA recovered can be used for any application that requires high purity RNA.

Use the MEGAclear lit to clean up any of the following:

• In vitro transcribed RNA:
  • RNA from MEGAscript reactions
  • Amino allyl-modified RNA
  • Biotinyled RNA
  • Cy dye labeled RNA
  • Capped RNA (e.g., from mMESSAGE mMACHINE kit reactions)
• Total RNA

Using the MEGAclear kit

The protocol is quick and easy. The transcription reaction is mixed with Binding Solution, passed over an RNA-binding filter, and washed; an elution step releases the purified RNA, which is now ready for any application requiring highly pure synthetic or in vitro transcribed RNA. For RNA recovery of 70% or greater, single-stranded RNA should be >100 nt and double-stranded RNA should be >200 bp.
