High Mobility Group Box 1 (HMGB1) recombinant human protein is supplied as a lyophilized powder. This protein is suitable for use in protein studies such as protein structure analysis and protein-protein interactions. In general, recombinant proteins can also be used as an immunogen, as a protein standard, or in cell biology research applications.
Background: Human High-Mobility Group Box 1 protein (HMGB1), also known as HMG-1 or amphoterin previously, is a member of the HMGB family consisting of three members, HMGB1, HMGB2 and HMGB3. HMGB1 is a nonhistone architectural chromosomal protein ubiquitously present in all vertebrate nuclei and binds double-stranded DNA without sequence specificity. HMGB1 is structurally composed of two homologous DNA-binding domains entitled box A and box B and a highly negative-charged C terminus. This protein is extremely conserved across species with 99- 100% aa sequence homology to mouse, rat, bovine, canine, and porcine proteins. As a nuclear protein, HMGB1 stabilizes nucleosomes and allows bending of DNA that facilitates gene transcription which is essential for individual survival. Meanwhile, it is revealed that HMGB1 can also act as a cytokine extracellularly and regulates monocyte, T cell, dendritic cell activities in inflammatory responses. Once secreted, HMGB1 induces signal transduction through its receptor RAGE and perhaps TLR2 and TLR4. The immune-regulatory function of HMGB1 may lead to being identified as new therapeutic target.
