Catalog No. P2996

The PKC family of serine⁄threonine kinases, including PRKCQ (PKC theta), is activated intracellularly by signal transduction pathways. In humans, at least 12 different PKC polypeptides have been identified. These isoforms differ in primary structure, tissue distribution, subcellular localization, mode of action in vitro, response to extracellular signals, and substrate specificity. PKC alpha, beta I, beta II and gamma form the conventional family; their activities are Ca2+- and phospholipid-dependent.

PRKCQ (PKC theta)
PRKCQ (PKC theta) was pre-diluted in enzyme dilution buffer and assayed in 20mM HEPES (pH 7.4), 10mM MgCl2, 0.1mM EGTA, 1X lipid mix (200μg/mL phosphatidylserine, 20μg/mL diacylglycerol, 1mM HEPES (pH 7.4), 0.03% CHAPS), 500μM ATP 100μg/mL PKC epsilon peptide substrate (ERMRPRKRQGSVRRRV) and trace [32P]-γ-ATP for 10 minutes at 30°C
Kinase and inhibitors
6,640nmoles of phosphate transferred to PKC epsilon peptide substrate (ERMRPRKRQGSVRRRV) per minute per mg of total protein at 30°C. Activity determined at a final protein concentration of 0.5 to 2μg/mL
His Tag
