Description

PIK3C3 (hVPS34) is the sole class III member of the PI3 Kinase family. PIK3C3 phosphorylates phosphatidylinositol (PI) to produce phosphatidylinositol 3-phosphate [PI(3)P], which recruits downstream effectors containing FYVE and Phox domains to membranes. PIK3C3 is involved in coupling nutrient signaling to mTOR and also plays a role in autophagy and intracellular trafficking.

PIK3C3 (hVPS34) was diluted in assay buffer and the activity determined in a 10μL Adapta™ Assay (PV5099) with 10μM ATP and 100μM PI:PS Lipid Kinase Substrate (PV5122) at room temperature for 1 hour. The reaction was stopped with a 5μL addition of EDTA and detection reagents. The final concentrations are 10mM EDTA, 2 nM Adapta™ Eu-anti-ADP Antibody and 10 nM Alexa Fluor™ 647 ADP Tracer. The plate was read after a 45 minute room temperature incubation
