MAP2K2 was assayed in a two stage coupled reaction involving phosphorylation of inactive MAPK1 (PV3314) by MAP2K2 followed by phosphorylation of MBP substrate by activated MAPK1. The enzymes were diluted in enzyme dilution buffer before adding to the reaction. In stage one, MAP2K2 (8.33μg/mL final concentration) is incubated with inactive MAPK1 (33.3μg/mL final concentration) in 50mM Tris (pH 7.5), 10mM MgCl2, 0.25mM EGTA, 2.5mM DTT, 0.1mM Na3VO4, 5mM β-glycerophosphate, 0.01% Triton™ X-100 and 200μM ATP for 30 minutes at 30°C. In stage two, 5μL of the above reaction mix was transferred to 25μL of a reaction mixture containing 50mM Tris (pH 7.5), 10mM MgCl2, 0.25mM EGTA, 2.5mM DTT, 0.1mM Na3VO4, 5mM β-glycerophosphate, 0.01% Triton™ X-100, 200μM ATP, 667μg/mL MBP substrate and trace [32P]-γ-ATP and incubated for 10 minutes at 30°C
