The IgE receptor plays a central role in allergic disease, coupling allergen and mast cell to initiate the inflammatory and immediate hypersensitivity responses that are characteristic of disorders such as hay fever and asthma. The allergic response occurs when 2 or more high-affinity IgE receptors are crosslinked via IgE molecules that in turn are bound to an allergen (antigen) molecule. A perturbation occurs that brings about the release of histamine and proteases from the granules in the cytoplasm of the mast cell and leads to the synthesis of prostaglandins and leukotrienes--potent effectors of the hypersensitivity response. The IgE receptor consists of 3 subunits: alpha, beta ., and gamma .; only the alpha subunit is glycosylated.Description: The MAR-1 monoclonal antibody reacts with the Fc epsilon Receptor I alpha subunit, an IgE-binding subunit lacking signal-transducing ability. Fc epsilon RI alpha is expressed on mast and basophil cells and is up-regulated by the presence of IgE. Fc epsilon RI alpha forms a tetrameric complex with one beta and two gamma subunits. The beta and gamma subunits possess immunoreceptor tyrosine-based activation motifs (ITAM). The Fc epsilon RI complex plays an important role in triggering IgE-mediated allergic reactions.Applications Reported: This MAR-1 antibody has been reported for use in flow cytometric analysis.Applications Tested: This MAR-1 antibody has been tested by flow cytometric analysis of mouse bone marrow-derived mast cells. This can be used at less than or equal to 0.25 μg per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.PE-eFluor™ 610 can be excited with laser lines from 488-561 nm and emits at 607 nm. We recommend using a 610/20 band pass filter (equivalent to PE-Texas Red™). Please make sure that your instrument is capable of detecting this fluorochome.Light sensitivity: This tandem dye is sensitive to photo-induced oxidation. Please protect this vial and stained samples from light.Fixation: Samples can be stored in IC Fixation Buffer (cat. 00-8222) (100 μL of cell sample + 100 μL of IC Fixation Buffer) or 1-step Fix/Lyse Solution (cat. 00-5333) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation. Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically.Excitation: 488-561 nm; Emission: 607 nm; Laser: Blue Laser, Green Laser, Yellow-Green Laser.Filtration: 0.2μm post-manufacturing filtered.
