Fixation: Samples can be stored in IC Fixation Buffer (cat. 00-8222) (100 µL of cell sample + 100 µL of IC Fixation Buffer) or 1-step Fix/Lyse Solution (cat. 00-5333) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation. Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically. FceR1 (Fc epsilonR1 alpha) is a subunit of the high affinity receptor of IgE. Fc epsilonR1 alpha is a tetrameric complex consisting of one alpha, one β and two additional subunits. The latter two are required for signal transduction activity. The Fc epsilonR1 complex plays an important role in triggering allergic responses, and the immunoglobulin epsilon receptor (IgE receptor) is the initiator of the allergic response. When two or more high-affinity IgE receptors are brought together by allergen-bound IgE molecules, mediators such as histamine, which are responsible for allergy symptoms, are released. The IgE receptor couples allergen and mast cells to initiate the inflammatory responses that are characteristic of disorders such as hay fever and asthma.
