3CLpro, also known as main protease (Mpro), and PLpro are crucial enzymes for processing viral polyproteins, and due to their role in the SARS-CoV-2 virus life cycle have been identified as a promising drug target for blocking viral replication. The homogeneous EnzChek 3CLpro and PLpro Assay kits are ideal for easily screening and quantifying inhibitors of 3CLpro and PLpro.

The EnzChek 3C-like 3CLpro and PLpro Assay kits contain a fluorogenic protease-specific substrate, SARS-CoV-2 3CLpro or PLpro, positive control inhibitor, and assay buffer. Simply add your inhibitor of choice. Protease-catalyzed hydrolysis releases the fluorescently labeled peptide, and protease activity can be directly quantified from the change in fluorescence signal using a microplate reader (ex: 360 nm, em: 460 nm).
