CD244.1, PerCP-eFluor™ 710, clone: C9.1, eBioscience™
Rat Monoclonal Antibody
Manufacturer: Life Technologies LS46244080
This gene encodes a cell surface receptor expressed on natural killer (NK) cells (and some T cells) that mediate non-major histocompatibility complex (MHC) restricted killing. The interaction between NK-cell and target cells via this receptor is thought to modulate NK-cell cytolytic activity. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.Description: The monoclonal antibody C9.1 reacts with mouse CD244.1. This receptor is expressed on NK cells, gamma delta T cells, some CD8+ T cells, basophils, and monocytes on most inbred mouse strains except C57BL/6 and C58, which express the CD244.2 allotype. CD244 is a CD2-related receptor belonging to the SLAM family that exists as two different isoforms with opposing functions. CD244 associates with CD48, which is expressed widely on hematopoietic cells, regulates NK cell proliferation, cytotoxicity, and cytokine production. However, the CD244/CD28 interaction between NK and other cell types inhibits NK cytotoxicity, which contributes to self-tolerance. CD244 may enhance or inhibit CD8+ T cell effector function depending on SAP levels. The C9.1 antibody has been reported to have antagonist activity on activated NK cells.Applications Reported: This C9.1 antibody has been reported for use in flow cytometric analysis.Applications Tested: This C9.1 antibody has been tested by flow cytometric anaylsis of BALB/c mouse splenocytes. This can be used at less than or equal to 0.125 μg per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.PerCP-eFluor™ 710 emits at 710 nm and is excited with the blue laser (488 nm); it can be used in place of PerCP-Cyanine5.5. We recommend using a 710/50 bandpass filter, however, the 695/40 bandpass filter is an acceptable alternative. Please make sure that your instrument is capable of detecting this fluorochrome.Fixation: Samples can be stored in IC Fixation Buffer (cat. 00-8222) (100 μL of cell sample + 100 μL of IC Fixation Buffer) or 1-step Fix/Lyse Solution (cat. 00-5333) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation. Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically.Excitation: 488 nm; Emission: 710 nm; Laser: Blue Laser.Filtration: 0.2μm post-manufacturing filtered.
