The monoclonal antibody X1AO2 recognizes human Brachyury, a 47kDa protein encoded by the T gene and functioning as a nuclear transcription factor. This transcription factor binds to specific DNA regions near the palindromic sequence TCACACCT (T-site) through a region in its N-terminus, called the T-box. Brachyury functions during embryogenesis to regulate midline development by establishing the anterior/posterior axis through the regulation of genes involved in mesoderm formation and differentiation. Brachyury is the primary mesoderm marker used to distinguish differentiation of embryonic stem cells into a mesenchymal stem cell lineage. Variations in the T gene in humans are associated with neural tube defects and chordomas. Expression of brachyury is also increased in advanced stage lung tumors and tumor cell lines.
