The GeneAmp High Fidelity PCR System contains an enzyme blend of AmpliTaq DNA Polymerase and a new thermo stable proofreading enzyme. This proprietary blend increases the fidelity of PCR 3-fold over Taq DNA Polymerase. The careful blending of these enzymes prevents truncated products resulting from misincorporated nucleotides, and limits primer degradation by the 3' to 5' exonuclease activity of the proofreader. The increased sensitivity of the unique GeneAmp High Fidelity Enzyme Mix enables detection down to 1 ng of complex templates such as human genomic DNA.
The system is supplied with buffers specifically designed to enhance fidelity, sensitivity, and yield of PCR experiments. Two versions of the buffer are included for flexibility. For ease of use, one version contains a final MgCl2 concentration of 1.5 mM. The second buffer does not contain MgCl2. A separate MgCl2 solution is also provided for optimizing MgCl2 concentration. The system allows for amplification of products ranging from a hundred bases up to 10 kb from human genomic DNA. It can be used with templates from various sources such as mammalian genomic DNA, artificial chromosomes, plasmids, and cDNA.
